PHARMACODYNAMIC PROPERTIES Colistin sulphate is an antibiotic belongs to the polymyxin-group and is identical to Polymixin E. Colistin is used for prevention and treatment of diseases caused by sensitive bacteria in rabbit, pig, poultry, cattle, sheep and goat. Colistin is primarily effective against gram-negative micro-organisms. Colistin (cationic polypeptides) interact with lipopolysaccharide (anionic) in outer membrane of bacteria and causes disorganisation of the bacterial cell membrane results in increased permeability of membrane, leakage and cell death. It also inhibits bacterial oxidative metabolism. The combination of these effects results in rapid bactericidal activity. Colistin also neutralises endotoxins produced by several gram-negative bacteria such as heat labile and heat stable endotoxins of enterotoxigenic E. coli and Salmonella spp.. Resistance to colistin is uncommon
PHARMACOKINETIC PROPERTIES Following in feed or water oral administration of therapeutic dose of 100,000 I.U. or 150,000 I.U. colistin sulphate/kg bw per day to pigs or chickens, respectively, no quantifiable blood levels of colistin could be determined. Due to the lack of significant absorption following oral administration, this antibiotic is very suitable to be applied in infections of the gastro-intestinal tract; such as E-coli, and salmonella infections. Little is known about the possible routes of metabolism of colistin. Inactivation of colistin could take place by the binding of colistin to intestinal phospholipids. When administered orally, colistin is excreted almost entirely via the faeces in its inactivated form; therefore possibility of environmental resistance is nil (green product).
CONTRA-INDICATIONS & OVERDOSING No contra-indication is implicated. No symptoms of overdosing have been reported in chickens and pigs when medicated by feed containing colistin sulphate up to 500,000 I.U./kg bw per day for 5 consecutive days.
SYNERGISTIC EFFECTS COLIVETo-4800S (PROMYCINE-4800®) can be combined with DOXYVETo-50S or with AMOXYVETo-50S. Note: Because DOXYVETo-50S and AMOXYVETo-50S are antagonist, COLIVETo-4800 should not be used with DOXYVETo-50S and AMOXYVETo-50S simultaneously.
